Characterization of cucumber mosaic virus and its satellite RNAs associated with tomato lethal necrosis in Serbia

A four-year survey (2012–2015) was carried out to examine the genetic diversity of cucumber mosaic virus (CMV) isolates infecting tomato, as well presence and their satellite RNAs (satRNAs), collecting a total 226 samples throughout most important growing regions in Serbia. Besides CMV-like symptoms, collected also exhibited more severe such systemic necrosis leaves, branches stems, accompanied by fruit malformation necrosis. In double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), CMV detected approximately one quarter tested samples. Identification confirmed conventional reverse transcription-polymerase chain reaction (RT-PCR) sequencing coat protein (CP) gene group 11 selected Serbian isolates. Phylogenetic analysis CP sequences these revealed heterogeneity, they fell into two different subgroups, IA II. An additional RT-PCR positive using specific primers satRNAs eight Sequence phylogenetic analyses showed that variants were very heterogeneous, belonging necrogenic non-necrogenic variants. Necrogenic divided groups, B B1, containing characteristic ‘necrogenic consensus’ sequence at 3? end RNA. necrotic phenotype co-determined expressed inoculated tomato plants.
